Polymerase chain reaction (PCR) is an experimental technique that uses polymerase enzymes to amplify specific genetic sequences that are flanked by a set of primers. PCR amplification can generate thousands of copies of a specific genetic sequence relatively rapidly, making it an ideal tool for identifying and studying specific mutations in the genetic code. PCR assay kits contain the reagents necessary to amplify a specific target sequence from genomic or plasmid DNA and rapidly identify the presence or absence of a well characterized mutant allele. PCR assay kits may use allele specific primers (recognizing only a specific mutant allele), or post-amplification RFLP (restriction fragment length polymorphism) to characterize the alleles present.
