Casilio HDR
Genomics-online offers plasmids for HDR (homology directed repair) mediated gene editing. HDR-mediated gene editing can be applied for the gene knock-out, correction of point mutations and introduction of beneficial mutations. Our plasmids use the Casilio system which combines CRISPR/dCas9 with a Pumilio RNA-binding domain to increase specificity. The Pumilio binding site (PBS) recruits RNA-binding protein-tethered DNA repair proteins (DRP) to target site via aptamers appended to guide RNA. Casilio enables multimerization of multiple DRP. This minimizes errors in DNA repair, a crucial factor in in possible fields of application such as gene therapy.
