Casilio Imaging
genomics-online offers plasmids in order to facilitate the fluorescent detection of DNA-binding proteins. The plasmids use the Casilio system which is an improvement to conventional CRISPR-Cas. Casilio is a hybrid system combining CRISPR/dCas9 and Pumilio RNA-binding domain. The distinctive feature are repositioned effector proteins which enables multiplexing and multimerization. By inserting multiple binding sites and thus recruiting multiple molecules fluorescent proteins, the signal-to-noise ratio of in experiments like chromosome labeling is significantly lowered. Where conventional systems would require multiple experiments with subsequent analytical integration, Casilio can perform analysis in a single experiment. Casilio-Imaging capitalizes on the multimerization capability to recruit 15 or more fluorescent proteins to a target as opposed to requiring target site tiling of dCas9-GFP direct fusions using 12 or more individual gRNAs in previous studies.
